Cognitive Judgement Bias Task

Also called the Ambiguity-Judgement Task or Pessimism Task, this assay tests the “glass half empty” phenomenon entirely through non-verbal audio tones or visual prompts. [10, 11]

Mechanism

The subject is trained to understand two extremes:

  • A high-pitched tone → “press a button to get a reward”
  • A low-pitched tone → “don’t press, or you will get a mild punishment” (time-out or annoying buzzer)

Once trained, researchers introduce an ambiguous, perfectly mid-pitched tone. [10, 11]

Depression Marker

  • Healthy children and animals display an “optimistic” bias — they press the button, anticipating a reward.
  • Depressed subjects show a “pessimistic” bias — they interpret the ambiguous tone as a threat or failure and refuse to act, mirroring the lack of motivation and fear of negative outcomes seen in clinical depression. [2, 12]

Drug Readout — Time-Course Differentiation

This task provides a critical temporal signature:

  • Rapid-acting antidepressants (e.g., ketamine) alter judgment instantly, causing an immediate shift toward the reward-predicting lever
  • Conventional antidepressants (e.g., fluoxetine, sertraline) only reverse pessimistic bias after chronic (2-3 weeks) administration

This provides an excellent metric for differentiating mechanisms and predicting human efficacy timelines in preclinical-drug-screening.
